CBSE Class 10 Science 2018 · Paper 31(B) · Question 24
In the experiment of preparing a temporary mount of a leaf peel to observe stomata, we use glycerine and safranin. State the role of the two liquids in this case.
Answer
Glycerine prevents the peel from drying; safranin stains the cells so that guard cells and stomata are seen clearly.
Glycerine: keeps the leaf peel moist and prevents it from drying up while it is observed under the microscope.
Safranin: a stain; it colours the cells of the peel (their walls and nuclei), so that the epidermal cells, the guard cells and the stomata become distinct and are seen clearly.
